File Server Capacity and Performance

I'm looking for some guidance on how to determine the capacity of a file
server. I know there are lots of variables that play in to the capacity.
Here are some that enter my mind:
# of users, # of open files, # of connections to a file share, # of shares,
speed of the disk subsystem (I/O), other services running on the server (AV,
quotas, etc), amount of data, size of the files, network performance,
running shadow copies, etc.

Does anyone know what the best way is to benchmark a file server and to
check the performance over time? Where should we look for pain points?
What is the best way to tune a file server? Is anyone using MS Server
Performance Advisor?

If I should post this in a different forum, let me know.
